Genetic Variability in Four Daylily Genus (Hemerocallis) Taxa using RAPD



Abstract
The potential use of RAPDs (randomly amplified polymorphic DNAs) to detect genetic polymorphism among sepecies was investigated using four species and one variety of dayliliy (Hemerocallis fulva Linne, H. fulva var, kwanso Regel, H. dumortieri Morr. and H. thunbergii Baker). With seventy polymorphic DNA bands which were amplified from these plants using 11 random primers, it was possible to identify a high level of intraspecific polymorphism among same H. fulva species, and interspecific polymorphism among different species investigated. A phenogram based on shared polymorphic amplification products confirmed the previous morphologically established relationship among the Hemerocallis species. Furthermore, it was possible to discriminate individual plants of same species which were not possible using morphological markers. Our results showed that H. dumortieri Morr. might be phylogenetically different from other Hemerocallis lines, and RAPD markers obtained might be useful for further genetic and breeding works.
